Podocyte Clusters

Podocyte clusters are organized groups of specialized epithelial cells that form the filtration barrier within developing kidney glomeruli, making them important for understanding renal development and function. During glomerular formation, precursor cells differentiate into podocytes, reorganize their actin-rich cytoskeleton, and extend interdigitating foot processes connected by slit diaphragms. This coordinated architecture supports selective filtration while maintaining the integrity of developing capillary loops. Studying podocyte clusters helps researchers examine cell differentiation, tissue organization, and signaling during nephron development, and provides a framework for investigating how disrupted podocyte structure contributes to proteinuria, glomerular disease, and potential regenerative approaches.

In this video, we describe the protocol to derive human kidney podocytes from induced pluripotent stem cells through cell differentiation under defined chemical conditions. The generated podocytes can be used for nephrotoxicity testing and disease modeling.

Single-channel Analysis and Calcium Imaging in the Podocytes of the Freshly Isolated Glomeruli

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

Spatial Separation of Molecular Conformers and Clusters

We present a technique that allows the spatial separation of different conformers or clusters present in a molecular beam. An electrostatic deflector is used to separate species by their mass-to-dipole moment ratio, leading to the production of gas-phase ensembles of a single conformer or cluster stoichiometry.

Cluster Sampling Method

2023

Appropriate sampling methods ensure that samples are drawn without bias and accurately represent the population. Because measuring the entire population in a study is not practical, researchers use samples to represent the population of interest. To choose a cluster sample, divide the population into clusters (groups) and then randomly select some of the clusters. All the members from these clusters are in the cluster sample. For example, if you randomly sample four departments from your...

Vesicular Tubular Clusters

2023

After budding out from the ER membrane, some COPII vesicles lose their coat and fuse with one another to form larger vesicles and interconnected tubules called vesicular tubular clusters or VTCs. These clusters constitute a compartment at the ER-Golgi interface known as ERGIC (Endoplasmic Reticulum Golgi Intermediate Compartment). The ERGIC is a mobile membrane-bound cargo transport system that sorts proteins secreted from ER and delivers them to the Golgi. With the help of motor proteins such...
