Spermatocyte Subpopulations

Spermatocyte subpopulations are distinct groups of male germ cells classified by developmental stage, molecular profile, or meiotic status, providing a framework for understanding sperm production. As spermatocytes progress through meiosis, they undergo DNA replication, homologous chromosome pairing and recombination, and successive divisions that generate genetically distinct germ cells; these events produce populations such as early and late primary spermatocytes and secondary spermatocytes. Characterizing these populations with cell morphology, gene-expression analysis, or single-cell approaches helps developmental biologists map spermatogenesis, identify stage-specific regulatory mechanisms, and investigate disruptions associated with infertility, reproductive disease, and abnormal germ-cell development.

Meiosis is the developmental process by which gametes are formed through a single round of DNA replication and two successive rounds of chromosome segregation. Mammalian meiosis can be examined by utilizing a technique to prepare meiotic chromosome spreads. Here, we demonstrate a method of preparing surface-spread nuclei from mouse spermatocytes.
