Tissue-scale Imaging

Tissue-scale imaging is the visualization of cells, structures, and dynamic events across intact tissues or tissue-sized specimens, bridging the gap between single-cell microscopy and whole-organism observation. It typically combines fluorescent labeling with optical sectioning, three-dimensional image acquisition, and computational reconstruction to capture spatial relationships while reducing the loss of information caused by physical sectioning. In developmental biology, these approaches reveal how cell division, migration, differentiation, and tissue remodeling are coordinated over time, enabling researchers to connect molecular signals with changes in tissue architecture. The resulting datasets support quantitative analysis of morphogenesis, organ formation, and developmental defects.

The protocol provides a detailed method of neuronal imaging in brain slice using a tissue clearing method, ScaleSF. The protocol includes brain tissue preparation, tissue clarification, handling of cleared slices and confocal laser scanning microscopy imaging of neuronal structures from mesoscopic to microscopic levels.

A methodology is described herein for representing anatomical imaging data within crystals. We create scaled three-dimensional models of biomedical imaging data for use in Sub-Surface Laser Engraving (SSLE) of crystal glass. This tool offers a useful complement to computational display or three-dimensionally printed models used within clinical or educational settings.
