Unicellular Tubulogenesis

Unicellular tubulogenesis is the process by which a single cell forms a hollow, tube-like structure, providing a model for understanding how biological tubes arise during development. It depends on coordinated cell polarization, targeted membrane trafficking, cytoskeletal remodeling, and controlled shaping or expansion of a lumen within the cell. Studying this process in developmental biology reveals how cells organize their membranes and internal architecture to generate specialized tubular compartments. These insights support research into epithelial morphogenesis, organ formation, and congenital disorders in which abnormal tube development disrupts tissue structure and function.

The C. elegans excretory canal is a unique single-cell model for the visual in vivo analysis of de novo polarized membrane biogenesis. This protocol describes a combination of standard genetic/RNAi and imaging approaches, adaptable for the identification and characterization of molecules directing unicellular tubulogenesis, and apical membrane and lumen biogenesis.
