Wolffian Duct Morphogenesis

Wolffian duct morphogenesis is the embryonic formation and remodeling of the nephric duct, an epithelial tube that supports urinary and reproductive system development. During organogenesis, ductal cells proliferate, migrate, polarize, and establish a continuous lumen, while signals such as GDNF-RET regulate ureteric bud outgrowth and interactions with surrounding mesenchyme guide branching and tissue patterning. Studying this process clarifies how epithelial tubes acquire precise architecture and connect developing organs. It also provides a framework for investigating congenital kidney and reproductive tract defects, sex-specific developmental pathways, and the cellular mechanisms that coordinate tissue growth, signaling, and morphogenesis.

Organ Culture and Whole Mount Immunofluorescence Staining of Mouse Wolffian Ducts

We present a method for isolation and culture of the mouse Wolffian duct (WD). We also demonstrate a detailed procedure for whole mount immunostaining of cultured/freshly isolated WDs with fluorescently tagged antibodies. Together, these techniques enable the study of WD development, coiling, and differentiation.

Cannulation of the Mouse Submandibular Salivary Gland via the Wharton's Duct

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.
