Non-invasive Model

A non-invasive model is a mathematical or computational representation that evaluates a system without physically penetrating, damaging, or interrupting it, making it valuable when direct testing is impractical or risky. It works by relating externally collected observations, such as measurements from sensors or imaging, to hidden states, material behavior, or system performance through equations, simulations, or data-driven methods. In engineering, these models support condition monitoring, fault diagnosis, performance prediction, and design validation while preserving the integrity and operation of the system. Their accuracy depends on measurement quality, appropriate assumptions, and validation against reliable experimental or operational data.

Murine Perineural Invasion Model: Generating an In Vivo Model to Simulate Perineural Invasion in Mouse Sciatic Nerve

2025

In this video, we develop an in vivo mouse model for perineural invasion by injecting pancreatic cancer cells into the sciatic nerve. This model helps quantify the extent of invasion and study the interactions between cancer cells and nerves.

Generation of a Three-Dimensional Spheroid Model of Tumor Cell Invasion

Source: Guyon, J., et al. A 3D Spheroid Model for Glioblastoma. J. Vis. Exp. (2020)The video demonstrates the generation of a three-dimensional spheroid model of tumor cell invasion. Spheroids are formed from human brain tumor cells and embedded in a collagen matrix. The tumor cells at the spheroid periphery adopt an invasive phenotype, secreting proteases to degrade the collagen and advancing to invade the surrounding matrix.

In this video, we describe the procedure to establish a murine model of perineural invasion by injecting cancer cells into the sciatic nerve. The model can help gain an understanding of the magnitude of nerve invasion for subsequent assessment of anti-cancer drugs.

Models of tumor cell invasion into three-dimensional extracellular matrix better reflect the in vivo situation than two-dimensional motility assays. Using matrix invasion assays combined with confocal imaging of fluorescently-labeled cells, detailed information on invasion modes and the distinct contributions of leading versus following cells can be obtained.
