Parabolic Stress Profile

A parabolic stress profile is a quadratic variation of stress across a material’s cross-section, commonly describing transverse shear stress in a rectangular beam. Under shear loading, the stress is zero at the outer surfaces and increases toward the neutral axis because the internal shear force is distributed unevenly across the section; for a rectangular section, the maximum value occurs at the center and equals 1.5 times the average shear stress. Engineers use this profile to evaluate beam strength, identify critical locations, and predict failure or deformation in structural components, shafts, plates, and other load-bearing designs.

Here, we describe a method to analyze changes in the initiation of mRNA translation of eukaryotic cells in response to stress conditions. This method is based on the velocity separation on sucrose gradients of translating ribosomes from non-translating ribosomes.
