Dissolved Oxygen Measurement

Dissolved oxygen measurement is the determination of oxygen concentration in water or other liquids, a key indicator of aquatic ecosystem health and water quality. Electrochemical sensors measure the current generated as oxygen is reduced at a cathode, while optical probes determine oxygen levels from oxygen-dependent quenching of a luminescent signal; both methods require calibration and careful control of temperature and pressure. Environmental scientists use these measurements to assess respiration, photosynthetic activity, organic pollution, and eutrophication, as well as to monitor wastewater treatment and conditions affecting fish and microbial communities. Reliable oxygen data supports pollution control, ecosystem management, and environmental change studies.

This article describes a simple and reproducible protocol to manipulate dissolved oxygen conditions in a laboratory setting for animal behavior studies. This protocol may be used in both teaching and research laboratory settings to evaluate organismal response of macroinvertebrates, fishes, or amphibians to changes in dissolved oxygen concentration.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
