Ethanol Preservation

Ethanol preservation is a method for stabilizing biological specimens or environmental samples by storing them in ethanol, which slows deterioration and maintains material for later analysis. Ethanol removes water and disrupts proteins and cell membranes, suppressing microbial growth and many enzymatic reactions; preservation quality depends on ethanol concentration, sample type, and storage conditions. In environmental research, this approach supports the collection and transport of invertebrates, tissue, and DNA-bearing material from field sites. Proper preservation can improve morphological identification and molecular analyses while reducing sample loss between field collection and laboratory processing.

2012

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Biofuels: Producing Ethanol from Cellulosic Material

2023

Source: Laboratories of Margaret Workman and Kimberly Frye - Depaul University In this experiment, cellulosic material (such as corn stalks, leaves, grasses, etc.) will be used as a feedstock for the production of ethanol. The cellulosic material is first pretreated (ground and heated), digested with enzymes, and then fermented with yeast. Ethanol production is monitored using an ethanol probe. The experiment can be extended to optimize ethanol production by varying the feedstock used,...

Source: Bucher, T., et al. Methodologies for Studying B. subtilis Biofilms as a Model for Characterizing Small Molecule Biofilm Inhibitors. J. Vis. Exp. (2016).This video demonstrates how biofilm matrix integrity influences bacterial resistance to ethanol by comparing colony-forming units in untreated and inhibitor-disrupted biofilms. Reduced CFUs in inhibitor-treated samples demonstrate that the extracellular matrix is essential for protecting biofilm-embedded cells from antimicrobial stress.
