Copper Growth Assay

A copper growth assay is a laboratory method that measures how cells grow when exposed to defined concentrations of copper, providing a functional readout of copper tolerance and homeostasis. In a typical experiment, genetically matched cultures are grown with increasing copper levels, and growth is quantified by comparing treated samples with untreated controls or reference strains. In genetics, the assay helps determine how mutations or gene-expression changes affect copper uptake, transport, sequestration, and detoxification, linking genotype to phenotype. It supports studies of metal-responsive pathways, gene function, and cellular adaptation to environmental stress.

Source: Kadoya, S., Sano, D. Assays for the Specific Growth Rate and Cell-binding Ability of Rotavirus. J. Vis. Exp. (2019).This video demonstrates the use of a plaque assay to assess the rotavirus growth curve. The method involves infecting host cells with diluted virus, overlaying with agarose to restrict spread, and staining to visualize plaques formed by viral replication. By counting plaque-forming units at defined time points, the resulting growth curve reveals the lag, exponential, and...

2025

In this video, we demonstrate the luciferase assay to quantify the growth of the intracellular pathogen Toxoplasma gondii. Utilizing a strain of Toxoplasma that expresses luciferase enzyme after infecting the host cells, the luciferase activity is detected in the infected cell culture to determine the successful reproduction of the parasite.
