Dcas9 Sam System

The dCas9-SAM system is a CRISPR-based gene-activation method that increases transcription without cutting DNA, making it valuable for studying gene function and regulation. It uses catalytically inactive Cas9, or dCas9, guided by a single-guide RNA to a selected promoter or regulatory sequence; RNA-binding MS2 aptamers then recruit a Synergistic Activation Mediator complex containing transcriptional activators such as VP64, p65, and HSF1. By stimulating endogenous gene expression at targeted loci, dCas9-SAM supports functional genomics, regulatory-network analysis, disease modeling, and investigations of cell-state control. Its programmable design also enables multiplexed activation of several genes in genetic research.

Antifouling Self-assembled Monolayers on Microelectrodes for Patterning Biomolecules

We present a procedure for forming a poly(ethylene glycol) self-assembled monolayer (PEG-SAM) on a silicon substrate with gold microelectrodes. The PEG-SAM is formed in a single step and prevents biofouling on silicon and gold surfaces. Electrophoresis is then used for patterning biomolecules down to the nanoscale.
