Mutant Colony Isolation

Mutant colony isolation is the process of separating and recovering microbial colonies that carry genetic changes from a mixed population, enabling researchers to study specific variants. After mutagenesis or genetic transformation, cells are spread on selective or differential media, where growth or colony appearance helps distinguish potential mutants; individual colonies are then picked, restreaked to establish clonal cultures, and verified using phenotypic assays or molecular analysis. In genetics, this workflow links genotype to observable traits and supports studies of gene function, regulatory pathways, metabolic variation, and resistance phenotypes. Reliable isolation provides stable, well-characterized strains for downstream experiments and reproducible comparisons.
