Faire Assay

The FAIRE assay (formaldehyde-assisted isolation of regulatory elements) is a genomic method for identifying regions of open chromatin, helping researchers map DNA sequences that may regulate gene expression. It uses formaldehyde to crosslink DNA to proteins, including nucleosomes; after cell lysis, sonication, and phenol-chloroform extraction, DNA not tightly associated with chromatin proteins preferentially partitions into the aqueous phase. Researchers can analyze the enriched DNA by PCR or sequencing to locate promoters, enhancers, and other regulatory elements. In genetics, FAIRE assay data supports studies of chromatin accessibility, developmental regulation, disease-associated variants, and genome-wide control of transcription.

Angiogenesis, the sprouting of blood vessels from pre-existing vasculature, is associated with both natural and pathological processes. Here we demonstrate an aortic ring assay that allows angiogenic potentiators and inhibitors to be directly added to aortic rings in culture. Sprouting and neovessel outgrowth can be determined by inspecting the aortic rings over a period of 6-12 days.
