Fragment Size Analysis

Fragment size analysis is a genetic method for determining the length of DNA fragments, providing information about sequence variation, molecular structure, and genetic identity. In gel or capillary electrophoresis, an electric field separates charged DNA molecules according to size, with shorter fragments migrating more rapidly through a porous medium; fluorescent labeling and size standards can support accurate measurement. Researchers use these profiles to assess PCR products, restriction digests, and short tandem repeat markers, enabling genotyping, mutation screening, linkage studies, sample identification, and quality control in molecular genetics. The results help distinguish alleles and verify that experimental products have the expected composition.

Leprosy, caused by Mycobacterium leprae, is still endemic in many places. In order to learn about the spread and mode of transmission of leprosy, it is important to determine which strain of M. leprae has infected a patient. Variable numbers of tandem repeats (VNTR) typing is one such method.
