Knock-in Rat Models

Knock-in rat models are genetically engineered rats that carry a precisely introduced DNA sequence or mutation at a defined genomic location, enabling researchers to study gene function in a whole-animal context. Typically, CRISPR-Cas9 or related genome-editing tools create a targeted DNA break, while a donor template guides repair and inserts the desired allele, such as a disease-associated variant, human gene sequence, or reporter. In genetics research, these models help link specific mutations to physiological traits, investigate disease mechanisms, evaluate therapeutic targets, and assess drug responses. Their controlled genetic changes provide a powerful complement to naturally occurring variants and broader transgenic approaches.

Creation of Reversible Cholestatic Rat Model

2011

Cholestasis is a clinical condition commonly encountered by both surgeons and gastroenterologists. Creation of a reversible cholestatic rat model can be challenging in view of the smaller size and unique hepatopancreatobiliary anatomy in rats. This video article demonstrates the creation of a reversible cholestatic model.
