Per2 Promoter Reporter

A Per2 promoter reporter is a genetic tool that measures activity of the Per2 gene promoter, a regulatory DNA region controlling transcription of a core circadian clock gene. In this system, the Per2 promoter is linked to a reporter gene, commonly luciferase, so promoter-driven transcription produces a detectable bioluminescent signal that rises and falls with cellular clock activity. Researchers use these reporters to monitor circadian rhythms in living cells or tissues, compare genetic or environmental effects on clock regulation, and analyze the timing and strength of transcriptional responses. The approach connects promoter function with measurable phenotypes in molecular genetics and chronobiology.

The Eukaryotic Promoter Region

2020

The eukaryotic promoter region is a segment of DNA located upstream of a gene. It contains an RNA polymerase binding site, a transcription start site, and several cis-regulatory sequences. The proximal promoter region is located in the vicinity of the gene and has cis-regulatory sequences and the core promoter. The core promoter is the binding site for RNA polymerase and is usually located between -35 and +35 nucleotides from the transcription start site. The distal promoter regions are...

A Lentiviral Transcriptional Reporter System to Generate a Glioblastoma Stem Cell Differentiation Reporter Line

2025

This video demonstrates a technique to generate a glioblastoma stem cell reporter line to identify the differentiation of glioblastoma stem cells (GSCs) into astroglia. It outlines the steps involved in seeding GSCs, transducing them with a lentivirus encoding GFP, and determining GFP reporter expression through flow cytometry.

Promotion Metrics

2025

Businesses rely on various promotional metrics to gauge the effectiveness of marketing campaigns. Baseline sales represent the average sales without any promotional efforts, while incremental sales measure the increase in sales resulting from a promotion. For example, if a product typically sells 300 units and a promotion increases sales to 400 units, the incremental sales are 100. This distinction is crucial for evaluating the true impact of promotional activities. Impressions count the number...
