Sirna-mediated Depletion

siRNA-mediated depletion is a gene-silencing method that reduces the abundance of a specific messenger RNA and its corresponding protein, enabling researchers to assess gene function. Introduced small interfering RNAs are incorporated into the RNA-induced silencing complex, whose guide strand pairs with a complementary target transcript; Argonaute then cleaves the RNA, promoting its degradation and limiting translation. In genetics, this approach supports targeted loss-of-function studies, pathway analysis, and validation of candidate gene roles in cellular phenotypes. Compared with permanent genome editing, siRNA-mediated depletion offers transient, adjustable suppression, making it useful for testing gene function and studying processes where reversible perturbation is important.

Here, we present a protocol for specific siRNA-mediated mRNA depletion followed by immunofluorescence analysis to evaluate meiotic spindle assembly and organization in mouse oocytes. This protocol is suitable for in vitro depletion of transcripts and functional assessment of different spindle and/or MTOC-associated factors in oocytes.
