Stem Cell Characterization

Stem cell characterization is the systematic assessment of a stem cell population’s identity, developmental potential, and genetic stability, helping researchers distinguish genuine stem cells from differentiated or abnormal cells. It combines analysis of cell-surface markers, gene-expression profiles, chromosome integrity, and functional behavior, including the capacity for self-renewal and differentiation into specialized cell types. In genetics, these measurements reveal how regulatory networks control cell fate and help identify mutations or epigenetic changes that alter development. Reliable characterization supports disease modeling, drug screening, regenerative medicine, and the quality control of cells used in research and therapeutic development.

The method described isolation and characterization of human Dental Pulp Stem Cells (hDPSCs) by using either enzymatic dissociation of pulp (DPSC-ED) or direct outgrowth of stem cells from pulp tissue explants (DPSC-OG). Then followed by in vitro comparative differentiation of both types of hDPSCs into odontoblasts.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
