Antibody Secretion Assay

An antibody secretion assay is a laboratory method that measures immunoglobulins released by antibody-producing cells, helping quantify humoral immune responses. In common formats, cells are cultured on a surface containing capture molecules; secreted antibodies bind nearby, and enzyme-linked or fluorescent detection reveals each responding cell as a spot or signal. In immunology and infection research, these assays identify antigen-specific B cells, compare responses after infection or vaccination, assess antibody production during disease, and evaluate immune therapies. Their single-cell resolution complements bulk antibody measurements, supporting studies of protective immunity, memory responses, and pathogen-specific humoral defense.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

This video demonstrates an imaging method to visualize in situ interferon-gamma (IFN-γ) secretion in the mouse spleen. Following bacterial infection and the induction of IFN-γ accumulation inside T cells and natural killer cells, the spleen is harvested from the mouse. Here, the methods for isolating human B cells from peripheral blood, differentiating human B cells into antibody (Ab)-secreting B cells (ASCs) in culture, and enumerating the total IgM- and IgG-ASCs via an ELISpot assay are described.
