Antibody Stripping

Antibody stripping is a laboratory technique that removes bound antibodies from biological samples so the same membrane or specimen can be analyzed with different antibodies. It typically uses combinations of low or high pH buffers, detergents, heat, or reducing agents to disrupt antigen–antibody interactions while preserving immobilized proteins and sample structure. In immunology and infection research, stripping supports sequential Western blotting and related assays, allowing researchers to examine multiple target proteins from one sample and improve comparisons across experimental conditions. Careful optimization is essential because harsh stripping conditions can damage epitopes, reduce signal quality, or affect subsequent antibody binding.

This protocol describes the development of two IgG class monoclonal antibodies (mAbs) strongly reactive to myoglobin of cetaceans. These mAbs are applied on a colloidal gold immunochromatographic test strip based on the sandwich format to differentiate the Mb of cetaceans from seal and other animals.
