Bacterial Survival Monitoring

Bacterial survival monitoring is the systematic measurement of whether bacterial populations remain viable over time, providing insight into infection dynamics and antimicrobial effectiveness. It works by exposing bacteria to defined conditions, such as immune components, host cells, or antibiotics, and tracking viable-cell abundance at selected time points using quantitative survival assays. Comparing bacterial recovery across conditions distinguishes persistence from clearance and reveals how environmental stress or immune pressure affects infection. In immunology and infection research, these measurements help evaluate host defense mechanisms, characterize bacterial persistence, and assess therapeutic responses, supporting the development of more effective antimicrobial strategies.

We describe a qualitative assay to monitor bacterial competition mediated by the Pseudomonas aeruginosa type VI secretion system (T6SS). The assay relies on the survival/killing of Escherichia coli target cells carrying a lacZ-reporter. This technique is adjustable to assess the bactericidal/bacteriostasis activity of T6SS-proficient microorganisms.

Bacteria may accumulate either detrimental or beneficial mutations during their lifetime. In a population of cells individuals that have accumulated beneficial mutations may rapidly outcompete their fellows. Here we present a simple procedure to visualize intraspecies competition in a bacterial cell population over time using fluorescently labeled...
