Binding Inhibition Analysis

Binding inhibition analysis is an assay-based approach for determining whether one molecule reduces the interaction between two binding partners, making it useful for characterizing molecular recognition in immunology and infection. In a typical competitive format, a labeled ligand, antigen, antibody, or receptor is mixed with increasing concentrations of an unlabeled inhibitor; reduced binding signal indicates competition, and the concentration producing a defined inhibition level can quantify inhibitory potency. Researchers use this analysis to compare antibody blocking activity, evaluate pathogen attachment inhibitors, characterize receptor-ligand interactions, and assess how mutations or treatments alter binding. The results support studies of immune recognition, infection mechanisms, and therapeutic development.

Biochemical reactions are occurring constantly in cells, converting starting substances to different products, usually with the help of enzymes that speed the reactions. Without enzymes, it would take far too long for most reactions to occur to be useful to the cell! Since enzymes help control the rate of reactions, their activity is regulated so that appropriate amounts of starting materials, intermediate metabolites, and products are maintained in the cell. Excessive build-up or depletion of...

Proteins bind to filamentous actin (F-actin) through distinct actin binding modules. In this video we demonstrate the procedure of actin co-sedimentation, which is an in vitro assay routinely used to analyze proteins or specific domains that bind F-actin.

Immunoblot Analysis of Apoptosis Inhibition in Host Cells Expressing an Antiapoptotic Bacterial Protein

Source: Berens, C. et al. Applying an Inducible Expression System to Study Interference of Bacterial Virulence Factors with Intracellular Signaling. J. Vis. Exp. (2015)This video demonstrates the use of immunoblotting to assess apoptosis inhibition by the bacterial effector protein CaeB in human cells. It outlines the steps for detecting apoptotic markers following doxycycline-induced expression of pro-apoptotic proteins in host cells expressing the effector.

Ligand Binding and Linkage

Allosteric proteins have more than one ligand binding site; the binding of a ligand to any of these sites influences the binding of ligands to the other sites. When a protein is allosteric, its binding sites are called coupled or linked. In the case of enzymes, the site that binds to the substrate is known as the active site and the other site is known as the regulatory site. When a ligand binds to the regulatory site, this leads to conformational changes in the protein that can influence the...
