Brain Tissue Penetrance

Brain tissue penetrance is the extent to which a drug, antibody, immune mediator, or infectious agent reaches the brain after entering the body, a key factor in treating and understanding central nervous system disease. Passage is governed largely by the blood-brain barrier, whose tight endothelial junctions, efflux transporters, lipid composition, and selective uptake systems restrict or facilitate movement from blood into neural tissue. Infection and inflammation can alter barrier permeability, potentially increasing access while also promoting edema and tissue injury. Measuring brain tissue penetrance supports antimicrobial and immunotherapy development, helps explain treatment failure in brain infections, and guides strategies for delivering effective concentrations to the central nervous system.

Inducing Penetrating Traumatic Brain Injury in Adult Drosophila Flies

This video demonstrates the induction of penetrating traumatic brain injury in the drosophila brain. Newly emerged flies are selected, anesthetized, and positioned under a fluorescence-equipped stereo microscope. The GFP-expressing mushroom body is identified, and a Minutien pin is inserted to cause injury. The flies are allowed to recover for further studies.

This video demonstrates the procedure to set up a recording chamber with a penetrating microelectrode array (PMEA) to capture neural signals from an unfolded hippocampus. This method allows us to study the speed and direction of propagation of neural activity within the unfolded hippocampus in vitro.
