Collagen Second Harmonic

Collagen second harmonic generation (SHG) is a label-free nonlinear optical signal that reveals the organization of collagen within biological tissues, making it valuable for studying extracellular matrix structure. In this process, two photons interact with the ordered, noncentrosymmetric arrangement of collagen fibrils and combine to produce one photon at twice the frequency, or half the wavelength, of the excitation light. In immunology and infection research, SHG microscopy enables real-time visualization of matrix remodeling, tissue injury, fibrosis, and changes in collagen architecture during immune responses or pathogen invasion. These measurements help link extracellular matrix organization with inflammation, disease progression, and tissue repair.
