Dextran Sedimentation

Dextran sedimentation is a cell-separation method that reduces the concentration of erythrocytes in whole blood to obtain a leukocyte-enriched suspension for laboratory analysis. The process uses dextran, a high-molecular-weight polysaccharide, to promote red blood cell aggregation and rouleaux formation, causing erythrocytes to settle under gravity while leukocytes remain in the upper plasma layer. In immunology and infection research, the resulting leukocyte-rich fraction supports immune-cell characterization, functional assays, pathogen-response studies, and downstream cell isolation. Its gentle, equipment-light approach can help prepare primary cells while preserving material for microscopy, flow cytometry, or culture-based experiments.
