Dna Tension Probes

DNA tension probes are molecular force sensors that measure the mechanical forces transmitted through cell-surface receptors, providing a way to study how physical cues regulate immune and infectious processes. Typically, a fluorophore-quencher DNA hairpin or mechanically labile duplex is tethered between a surface and a ligand; receptor-generated traction force opens or separates the DNA structure, producing a fluorescence signal proportional to force or force-dependent molecular events. In immunology, these probes can map forces exerted by T-cell receptors, integrins, or phagocytic machinery during recognition and activation. They also help clarify how pathogens and host cells use mechanical interactions during adhesion, entry, and immune evasion.

DNA stable-isotope probing is a cultivation-independent method to identify and characterize active communities of microorganisms that are capable of utilizing specific substrates. Assimilation of substrate enriched in heavy isotope leads to incorporation of labelled atoms into microbial biomass. Density gradient ultracentrifugation retrieves labelled DNA for downstream molecular analyses.
