Two-step Multiplex Nested Pcr

Two-step multiplex nested PCR is a molecular technique that amplifies and detects multiple genetic targets with enhanced sensitivity and specificity, making it useful for identifying infectious agents or immune-related markers. In the first PCR, several primer pairs simultaneously amplify distinct regions; a second PCR then uses primers located within the initial products to re-amplify selected targets and reduce nonspecific detection. This approach can reveal low-abundance nucleic acids in complex clinical or biological samples and support pathogen identification, co-infection analysis, and characterization of genetically related organisms. Its multiplex capacity also conserves sample material while expanding the information obtained from a single assay.
