Helper T Cell Lineages

Helper T cell lineages are specialized populations of CD4+ T lymphocytes that coordinate adaptive immune responses by directing other immune cells through targeted cytokine signaling. After recognizing antigen presented by major histocompatibility complex class II molecules, naïve CD4+ T cells differentiate in response to cytokine environments and lineage-defining transcription factors, generating subsets such as Th1, Th2, Th17, and regulatory T cells. Each lineage supports distinct functions, including macrophage activation, B cell help, mucosal defense, or immune suppression. Understanding these pathways clarifies host responses to infection and informs research on vaccines, inflammation, autoimmunity, and immune-based therapies.

Immunological Synapse Formation Between Helper T Cells and Antigen-Presenting Cells

2025

This video demonstrates the in vitro study of immunological synapse formation between an antigen-presenting cell and helper T lymphocytes using a fluorescence microscope. T cell receptors of helper T lymphocytes interact with an antigen-MHC-II complex in antigen-presenting cells and form immunological synapses, followed by secretory vesicle movements toward the synapse, which is visualized by fluorescent microscopy.

An In Vitro Technique to Differentiate GM-CSF Producing T Helper Cells from Naïve T Cells

2025

This video demonstrates the in vitro differentiation of granulocyte-macrophage-colony-stimulating factor or GM-CSF-producing T helper cells (THGM). Isolated naïve T cells are stimulated for differentiation into THGM, which is confirmed by detecting high intracellular GM-CSF production and a low expression of other T helper cell-specific cytokines.

Here, we present a protocol for a mosaic labeling technique that permits the visualization of neurons derived from a common progenitor cell in two distinct colors. This facilitates neural lineage analysis with the capability of birth-dating individual neurons and studying gene function in the same neurons of different individuals.
