Hla-dr Restricted Epitopes

HLA-DR-restricted epitopes are peptide sequences presented by HLA-DR, a class II major histocompatibility complex molecule, for recognition by CD4+ T cells. Antigen-presenting cells internalize proteins, digest them within endosomal compartments, and load resulting peptides into the HLA-DR binding groove; the peptide-HLA-DR complex then engages a specific T-cell receptor and can initiate cytokine production or other immune responses. Identifying these epitopes helps characterize host immunity to infectious agents, map antigen-specific T-cell responses, and guide vaccine design, immune monitoring, and immunotherapy. Their study also clarifies how pathogen-derived proteins are processed and presented during infection.

Mismatches in human leukocyte antigen (HLA) sequences between organ donor and recipient pairs are the major cause of antibody-mediated rejection in organ transplantation. Here we present the use of custom antigen arrays that are based on individual donors' HLA sequences to probe anti-donor HLA alloantibodies in organ recipients.

This video demonstrates an assay to detect anti-donor HLA alloantibodies in organ recipients. A peptide array is synthesized to represent potential epitopes for antibody-mediated organ rejection. The array is incubated with the recipient's serum, containing alloantibodies that bind to their target epitopes. Antibody binding is detected using chemiluminescence to identify donor-specific HLA epitopes that incite alloantibody reactivity.

Characterizing T-cell epitopes of pathogens that cause localized infections such as human papillomavirus is a challenge because of limited number of T cells in circulation. A method is described in which rare T cells were isolated and were characterized starting with a very small number of cells.
