Infection Driven Immune Modulation

Infection-driven immune modulation is the process by which invading pathogens alter the activity, coordination, and persistence of host immune responses, shaping the outcome of infection. Pathogen-associated molecular patterns are detected by pattern-recognition receptors, which trigger signaling networks that regulate cytokines, interferons, leukocyte recruitment, inflammation, and antimicrobial defenses; microbes can also redirect these pathways to evade clearance or promote tissue damage. Understanding this interplay helps explain why infections produce distinct inflammatory phenotypes and why immunity may become protective, dysregulated, or suppressed. In immunology and infection research, it informs vaccine design, anti-infective therapies, immunomodulatory treatments, and strategies for limiting immunopathology while preserving pathogen control.

2015

Drosophila melanogaster is an outstanding model organism for studying innate immune systems and the physiological consequences of infection and disease. This protocol describes how to deliver robust and quantitatively repeatable bacterial infections to D. melanogaster, and how to subsequently measure infection severity and quantify the host immune response.

Evaluation of Vaccine-Induced Immunity Against Bacterial Infection in a Mouse Model

2025

In this video, we describe a protocol to test the efficacy of an acellular pertussis vaccine against Bordetella pertussis infection in mice. Post-vaccination, the mice were subjected to infection, and their tissue homogenate was cultured to analyze the bacterial colonies as a readout of the vaccine-induced protective immunity.

Assessing Impaired MHC-I Antigen Presentation in Immune Cells Infected with Bacteria

Start with an immune cell culture infected with pathogenic bacteria that form intracellular inclusion bodies. The bacterial infection impairs the antigen presentation pathway in the immune cells.Take an uninfected culture as a control.Next, add a model protein antigen and incubate.Immune cells internalize, process, and present the model antigen-derived peptides on MHC-I molecules. After incubation, wash the cells to remove any unprocessed antigen.Fix the cells, then add a quenching agent to...
