Intracellular Bacteria Quantification

Intracellular bacteria quantification is the measurement of bacterial burden within host cells, providing a key readout of infection, survival, and replication. A common approach uses a gentamicin protection assay, in which the antibiotic eliminates extracellular bacteria while bacteria inside host cells remain protected; researchers then lyse the cells and quantify recovered organisms by colony formation or complementary molecular methods. In immunology and infection research, this technique helps evaluate bacterial entry, intracellular persistence, antimicrobial activity, and the effects of host immune responses. Combining culture-based counts with microscopy or genetic detection can clarify how pathogens interact with different cell types and treatments.

Here, we describe a phenotypic assay applicable to the High-throughput/High-content screens of small-interfering synthetic RNA (siRNA), chemical compound, and Mycobacterium tuberculosis mutant libraries.
