Lymphocyte Populations

Lymphocyte populations are the distinct groups of lymphocytes, including B cells, T cells, and natural killer cells, that coordinate adaptive and innate immune defenses. They arise from common lymphoid progenitors and acquire specialized receptors, surface markers, and effector functions through differentiation, antigen recognition, clonal expansion, and regulated communication with other immune cells. Characterizing these populations helps researchers distinguish naïve, memory, regulatory, cytotoxic, and antibody-producing cells during infection and vaccination. Population changes measured by flow cytometry, cell sorting, or molecular profiling can reveal immune activation, dysfunction, and disease progression, supporting studies of vaccines, immunodeficiency, autoimmunity, and immunotherapies.

Isolation and Th17 Differentiation of Naïve CD4 T Lymphocytes

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.

An In Vitro Technique to Stimulate Lymphocytes via Pathogenic Bacteria

This video demonstrates an in vitro technique to stimulate lymphocytes via non-typeable Haemophilus influenzae (NTHi) and to assess the immune response. Incubating isolated peripheral blood mononuclear cells (PBMCs) with NTHi causes the bacterial cells to stimulate T lymphocytes, leading to cytokine production. Upon inhibiting the secretion of cytokines, immunofluorescence staining is performed to detect the accumulated intracellular cytokines in stimulated T lymphocytes.

Lymphocytes are the major players in adaptive immune responses. Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.

The video demonstrates an immunological method to monitor in vivo adjuvant-induced cytotoxic T lymphocyte or CTL production. A mouse is pre-injected with genetically engineered, proliferative dye-stained donor CD8+ T lymphocytes. Subcutaneous injection of ovalbumin with adjuvants generates cytotoxic CD8+ T lymphocytes that are identified post-harvesting through immunostaining and flow cytometry.
