Microscopy Imaging

Microscopy imaging is a set of methods for forming magnified, resolved images of cells, tissues, microorganisms, and molecular structures, making otherwise invisible biological organization observable. It works by directing light or electrons onto a specimen and capturing transmitted, reflected, or emitted signals, with contrast enhanced through labels such as fluorescent probes or antibodies. In immunology and infection research, these approaches reveal pathogen localization, immune-cell interactions, tissue damage, and changes in cellular morphology over time. Imaging therefore connects molecular recognition with spatial and temporal behavior, supporting infection diagnosis, host-pathogen studies, vaccine research, and evaluation of antimicrobial or immunomodulatory treatments.

This article describes a working protocol to image dendritic spines from hippocampal neurons in vitro using Structured Illumination Microscopy (SIM). Super-resolution microscopy using SIM provides image resolution significantly beyond the light diffraction limit in all three spatial dimensions, allowing the imaging of individual dendritic spines with improved detail.

Source: Peiman Shahbeigi-Roodposhti and Sina Shahbazmohamadi, Biomedical Engineering Department, University of Connecticut, Storrs, Connecticut Optical microscopes have been around for centuries, and while they reached their theoretical limitation of resolution decades ago, new equipment and techniques, such as confocal and digital image processing, have created new niches within the field of optical imaging. The best optical microscopes will typically have a resolution down to 200 nm in ideal...
