Monocyte Activity

Monocyte activity is the set of immune functions performed by circulating monocytes, white blood cells that help connect rapid innate defense with longer-term tissue responses. After sensing pathogen-associated or damage-associated molecular patterns through pattern-recognition receptors, monocytes adhere to activated endothelium, migrate into tissues, and can differentiate into macrophage-like or dendritic-cell-like populations. They then contribute through phagocytosis, antigen processing, and secretion of cytokines and chemokines that recruit and activate other immune cells. In immunology and infection research, measuring monocyte activation helps clarify host-pathogen interactions, inflammatory disease mechanisms, immune suppression, and responses to vaccines or antimicrobial treatments.

Visualization of Caspase Activation in Human Monocyte-Derived Macrophages

This video demonstrates the activation of caspase proteins in human macrophages using bimolecular fluorescence complementation reporter proteins. The pro-domains of caspase-1 are fused with non-fluorescent fragments of Venus proteins, which are recruited to inflammasome complexes. The proximity of inflammasomes induces the refolding and fluorescence of Venus fragments, confirming caspase activation in the macrophages.

Activation and Measurement of NLRP3 Inflammasome Activity Using IL-1β in Human Monocyte-derived Dendritic Cells

Dendritic cells (DCs) secrete IL-1β in response to TLR8 recognition of synthetic purine, R848, followed by NLRP3 inflammasome activation with nigericin, therefore, IL-1β can be used to measure NLRP3 inflammasome activity. Intracellular cytokine staining, immunoblotting, and ELISA are used to accurately measure NLRP3 inflammasome priming and activation via IL-1β expression.

Here we present a protocol to quantify the chemotactic activity of blood monocytes in mouse models, to assess the effects of nutritional, pharmacological and genetic interventions on blood monocyte and to characterize the blood monocytes derived macrophages in mouse models using monocyte-chemoattractant protein-1 (MCP-1)-loaded basement membrane-derived gel plugs.
