Nasopharyngeal Sample Processing

Nasopharyngeal sample processing is the laboratory handling of specimens collected from the upper respiratory tract to preserve, prepare, and analyze material relevant to infection and immunity. After a swab is placed in transport medium, mixing releases mucus, epithelial cells, leukocytes, and microorganisms; controlled aliquoting, centrifugation, or clarification can separate particulate material before nucleic-acid extraction, antigen testing, or cellular and cytokine analysis. In immunology and infection research, consistent processing helps maintain sample integrity, reduce inhibitors and contamination, and support reliable detection of respiratory pathogens alongside measurements of local host responses. These workflows underpin diagnostic testing, surveillance, and studies of mucosal immunity during infection or after vaccination.

Methods to examine contributions of the nasopharyngeal-associated lymphoreticular tissues (NALT) to nasal and systemic immune responses of mice to intranasal vaccines are described. We demonstrate a surgical procedure to establish a NALT-dependent mouse model and ex vivo cultures of extracted NALT.
