Neutrophil Ros Production

Neutrophil ROS production is the generation of reactive oxygen species by neutrophils, white blood cells that provide rapid defense against invading pathogens. During the oxidative burst, activation of the NADPH oxidase complex transfers electrons to oxygen, producing superoxide and related oxidants within phagosomes and at the cell surface. These reactive molecules damage microbial membranes, proteins, and nucleic acids, working alongside phagocytosis, antimicrobial granules, and neutrophil extracellular traps to control infection. Because excessive or misdirected ROS can injure host tissues, studying this process helps explain both protective immunity and inflammatory disease, while informing research on immunodeficiency, infection outcomes, and therapies that regulate neutrophil activity.

This video demonstrates the induction and visualization of neutrophil extracellular traps or NETs in pre-labeled neutrophils using patient serum containing anti-neutrophil cytoplasmic antibodies. Furthermore, using an impermeable DNA dye and fluorescence microscopy enables the visualization of NETs.

ROS Detection with DCFH-DA Staining: Measuring Total ROS in Colorectal Cancer Cell Lines

2023

This video describes the protocol for detecting total cellular reactive oxygen species (ROS) using 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA). This method helps visualize ROS localization in the adherent cells using a fluorescence microscope and further quantify ROS intensity with a fluorescence plate reader.

An In Vitro Assay to Study the Interaction between Neutrophils and Biofilm

2025

This video demonstrates an in vitro assay to study neutrophil-biofilm interaction. The pathogenic bacteria embedded in the biofilm and isolated neutrophils were tagged using fluorescent labels, and the immune response by the neutrophils and the evasion of the immune response by the bacteria were assessed using a fluorescence microscope.
