Nlrp3 Inflammasome

The NLRP3 inflammasome is an intracellular immune-signaling complex that detects cellular stress and microbial threats, helping initiate inflammation during infection and tissue injury. After activation by diverse signals, NLRP3 assembles with the adaptor ASC and procaspase-1, enabling caspase-1 activation, maturation of interleukin-1β and interleukin-18, and inflammatory cell death known as pyroptosis. In immunology and infection research, studying this pathway clarifies how innate immune cells respond to pathogens and harmful stimuli. Its regulation is important for understanding host defense, chronic inflammatory disease, and potential therapies that selectively limit excessive inflammasome activity.

Detection of Inflammasome Activation via Fluorescence Microscopy

This video demonstrates a method to detect NLRP3 inflammasome activation in macrophages using fluorescent reporter probes. The cells are treated with potassium ionophores and glycine to induce inflammasome assembly and caspase-1 activation. Fluorescent reporter probes visualize activated caspase-1, and cells are stained with a fluorescent dye for nuclei visualization.

Dendritic cells (DCs) secrete IL-1β in response to TLR8 recognition of synthetic purine, R848, followed by NLRP3 inflammasome activation with nigericin, therefore, IL-1β can be used to measure NLRP3 inflammasome activity. Intracellular cytokine staining, immunoblotting, and ELISA are used to accurately measure NLRP3 inflammasome priming and activation via IL-1β expression.

We describe the detection of NLRP3 inflammasome activation on cellular basis using fluorescence microscopy and staining for active caspase-1 and the adaptor, ASC. A lactate dehydrogenase release assay is presented to detect pyroptotic lysis on a population basis. These techniques can be adapted to study many aspects of inflammasome biology.
