Ocular Microbiome Analysis

Ocular microbiome analysis is the study of microorganisms and their genetic material on the eye’s surface, especially the conjunctiva and cornea, to understand how microbial communities relate to ocular health and infection. Researchers typically collect tear or swab samples and use culture-independent methods such as DNA extraction, 16S rRNA gene sequencing, or metagenomic sequencing to identify microbial taxa and compare community composition, diversity, and abundance. In immunology and infection research, these data help clarify interactions between resident microbes and local immune defenses, distinguish healthy from disease-associated communities, and evaluate how antibiotics, inflammation, or other conditions alter the ocular environment, supporting more targeted diagnostics and therapies.

Changes in the oral microbiome throughout childhood are of growing interest. Comparison of different microbiome studies reveals a lack of standardized sampling protocols. Limited space makes sampling the sound subgingival sulcus of children challenging. Paper point sampling is presented here in detail as the method of choice for this area.

A Method to Induce Ocular Surface Inflammation in a Murine Model

This video demonstrates a method to induce ocular surface inflammation and meibomian gland dysfunction in a murine model. Stimulation of the mouse's immune system using an immunogen and later restimulation with the same immunogen promotes ocular surface inflammation, which causes plugging of meibomian gland secretion through neutrophil-produced aggNETs and leads to dry eye disease.

Experimental Glaucoma Induced by Ocular Injection of Magnetic Microspheres

We present a method for inducing elevated intraocular pressure (IOP), by injecting magnetic microspheres into the rat eye, to model glaucoma. This leads to strong pressure rises, and extensive neuronal death. This protocol is easy to perform, does not require repeat injections, and produces stable long-lasting IOP rises.
