Phagocyte Homogenization

Phagocyte homogenization is a laboratory method that disrupts phagocytic immune cells, such as macrophages and neutrophils, to produce a uniform cell lysate for analysis. Mechanical force, often combined with an appropriate lysis buffer, breaks the plasma membrane and releases intracellular proteins, enzymes, nucleic acids, and engulfed microbial material while helping preserve target components under controlled conditions. In immunology and infection research, the resulting homogenate supports investigations of pathogen uptake, intracellular killing, inflammatory signaling, and phagocyte function through biochemical, molecular, or proteomic assays. Careful control of disruption conditions improves sample consistency and the reliability of downstream measurements.

We present a high-throughput flow cytometric assay to determine the phagocytic activity of antigen-specific antibodies from clinical samples, utilizing fluorescent antigen-coated beads and a monocytic cell line expressing multiple Fc receptors—providing receptor usage and phagocytic activity determinations in a standardized and reproducible fashion for any antigen of interest.
