Rpe Cell Function

Retinal pigment epithelial (RPE) cell function encompasses the activities by which a specialized monolayer sustains photoreceptors, maintains the outer blood-retina barrier, and preserves retinal homeostasis. RPE cells transport nutrients and waste, recycle visual-cycle components, and phagocytose shed photoreceptor outer segments while regulating local fluid and immune conditions. In immunology and infection research, their barrier properties and production of cytokines and other signaling molecules help shape innate immune responses to tissue stress or invading pathogens. Defining how these functions change during infection or inflammation clarifies mechanisms of retinal injury and supports studies of ocular disease, host defense, and potential therapeutic strategies.

In this video we describe a technique to isolate iris and retinal pigment epithelial cells from porcine eyes to obtain their primary cultures for in vivo and ex vivo studies. These pigment cells could be genetically modified to study regenerative approaches to treat ocular disorders.

Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
