Spleen Weight Measurement

Spleen Weight Measurement is a quantitative method for assessing changes in spleen mass, an important indicator of systemic immune activity and disease-related effects. The procedure typically involves removing the spleen under controlled experimental conditions, removing surrounding tissue and excess fluid, and recording its mass with a calibrated balance, often alongside body-weight normalization. In immunology and infection research, increased spleen weight can reflect lymphoid expansion, inflammatory responses, or immune-cell accumulation during pathogen exposure, while reduced weight may indicate tissue damage or impaired immune function. These measurements support comparisons among experimental groups and help evaluate disease progression, treatment effects, and host responses.

A Mouse Model of Vascularized Heterotopic Spleen Transplantation

2025

The video demonstrates a surgical method for generating a mouse model of vascularized heterotopic spleen transplantation. The abdominal cavity of an anesthetized donor mouse is surgically opened to remove the spleen. The inferior vena cava and infrarenal aorta are identified, their lumbar branches are ligated, and openings are made for the heterotopic transplantation of a spleen from a congenic donor mouse. Upon completing vascular anastomosis and restoring blood flow to the implanted spleen,...

Assessing Bacterial Burden in the Mouse Spleen for Infection Studies

(2016)This video demonstrates the process of quantifying bacterial load in the spleen of an infected mouse to assess the severity of infection. The homogenized spleen is serially diluted and plated to determine total bacterial burden. The resulting data provide insights into host-pathogen interactions within...

Source: Ahn, J. J., et al. Experimental Infection with Listeria monocytogenes as a Model for Studying Host Interferon-γ Responses. J. Vis. Exp. (2016)The video demonstrates bacterial load measurement in spleen and liver tissues isolated from mice infected with an intracellular bacterial pathogen. Bacteria, accumulated within tissue-resident phagocytes during systemic infection, are released using detergent-based lysis and mechanical disruption. Serial dilutions of the homogenates are plated on...
