Splenocyte Isolation

Splenocyte isolation is the laboratory process of obtaining individual cells from the spleen to study immune function, inflammation, and host responses to infection. It typically involves mechanically dissociating spleen tissue into a cell suspension, removing debris and unwanted cell populations when needed, and maintaining conditions that preserve cell viability and activity. The resulting preparation contains diverse immune cells, including lymphocytes, macrophages, and dendritic cells, enabling flow cytometry, cell culture, functional assays, and pathogen-response studies. In immunology and infection research, reliable isolation supports analysis of immune-cell composition, cytokine production, antigen responses, and treatment effects.

2025

This video demonstrates an imaging-based method to visualize transplanted luciferase-expressing beta cells into a non-obese diabetic immunodeficient mouse. The introduction of splenocytes from a diabetic mouse leads to progressive beta cell destruction, visually confirmed by declining luminescence after substrate introduction.

Source: Tonya J. Webb1 1 Department of Microbiology and Immunology, University of Maryland School of Medicine and the Marlene and Stewart Greenebaum Comprehensive Cancer Center, Baltimore, Maryland 21201 ELISPOT is a standardized, reproducible assay used to detect cellular immune responses. The assay utilizes an enzyme-linked immunosorbent assay (ELISA)- based method to detect single-cell immune responses which can be visualized by spots, hence the name ELISPOT. ELISPOT was first described in...
