Spytag Spycatcher

SpyTag-SpyCatcher is a protein-labeling system that irreversibly links selected proteins, making it useful for studying molecular interactions in immunology and infection. It works when the short SpyTag peptide recognizes the SpyCatcher protein domain and forms a spontaneous covalent isopeptide bond between them, without requiring additional enzymes. Researchers can use this modular pairing to attach antigens to carrier proteins, organize proteins on nanoparticles, or immobilize recombinant molecules for assays. These applications support vaccine design, antigen presentation studies, pathogen interaction research, and the development of stable protein-based tools for investigating immune recognition.

This video demonstrates a method to generate multivalent-displaying outer membrane vesicle (OMV) vaccines. Mammalian cells are transfected to express and secrete viral antigens fused to a SpyTag and a polyhistidine tag.
