Tumor Antigen-specific T Cells

Tumor antigen-specific T cells are lymphocytes that recognize molecular markers displayed by cancer cells, making them important effectors of adaptive antitumor immunity. Their T-cell receptors bind tumor-derived peptide antigens presented by major histocompatibility complex molecules, activating signaling pathways that can drive cytokine production, target-cell killing, and coordination of broader immune responses. These cells can arise naturally in patients or be isolated, expanded, and selected for adoptive cell therapy, including approaches that enhance recognition of tumor-specific or tumor-associated antigens. Studying their specificity, persistence, and functional activity supports cancer immunotherapy development, biomarker research, and efforts to improve treatment precision while limiting damage to healthy tissues.

This video demonstrates an in vitro assay to validate chimeric antigen receptor (CAR) T cell activity. B-cell lymphoma cells expressing CD19 antigen and cytoplasmic luciferase are co-cultured with CD19-specific CAR T cells. CAR binding to CD19 on a tumor induces the CAR T cells to release cytokines and toxins that cause tumor cell cytotoxicity. The CAR T cell activity is evaluated by quantifying the released cytokine and decreased luciferase-mediated bioluminescence in the tumor cells.
