Tumor Antigen Targeting

Tumor antigen targeting is an immunological strategy that directs immune responses or therapeutic agents toward molecules expressed on cancer cells, helping distinguish malignant tissue from healthy tissue. It works by using antibodies, T-cell receptors, or engineered immune cells to recognize tumor-associated or tumor-specific antigens and trigger mechanisms such as immune-cell activation, direct killing, or delivery of cytotoxic treatment. In immunology and infection research, this approach supports the development of cancer vaccines, monoclonal antibodies, and adoptive cell therapies. Its effectiveness depends on antigen expression, tumor heterogeneity, immune access, and the ability to limit damage to normal tissues.

2019

This protocol describes a detailed workflow for the generation and ex vivo characterization of oncolytic viruses for expression of immunomodulators, using measles viruses encoding bispecific T cell engagers as an example. Application and adaptation to other vector platforms and transgenes will accelerate the development of novel immunovirotherapeutics for clinical translation.

An Ex Vivo Technique to Generate Tumor-Specific Chimeric Antigen Receptor T Cells

This video demonstrates a technique for the ex vivo generation of tumor antigen-specific chimeric antigen receptor (CAR) T cells. Retroviral vectors carrying transgenic RNA are mixed with spleen cells in a medium supplemented with interleukin-2 to promote T-cell survival and proliferation. The mixture is then transferred to a plate coated with recombinant human fibronectin fragments and centrifuged to facilitate the fusion of the virus with the cell, allowing the RNA to integrate into the host...

2023

This video describes the technique of pretargeted radioimmunotherapy or PRIT, in which immunoconjugates followed by radioligands are administered for treating tumors. This radioimmunotherapy reduces radiation doses to healthy tissues and utilizes radionuclides with very low half-lives incompatible with antibody-based vectors.
