Viral Particle Tracking

Viral particle tracking is a microscopy-based approach for following individual virions as they move through cells or tissues, helping researchers understand how infection begins and progresses. In this technique, viral particles are labeled with fluorescent markers and recorded over time, allowing single-particle tracking to reconstruct trajectories, measure movement, and identify interactions with cellular structures. In immunology and infection research, these measurements can reveal how viruses attach to receptors, enter host cells, traffic through intracellular compartments, and reach sites of replication. The resulting spatial and temporal data support studies of viral pathogenesis, host defenses, antiviral mechanisms, and strategies for improving therapeutic delivery.

Particle-tracking microrheology investigates the viscoelasticity of materials. Here, the technique is used to determine the viscoelasticity, creep compliance and effective crosslinking roles of different matrix components of a bacterial biofilm. The matrix consists of polymeric substances secreted by the bacteria and its components determine biofilm structure and mechanical properties.

We present an in vitro, two-color fluorescence assay to visualize the fusion of single virus particles with a fluid target bilayer. By labeling viral particles with fluorophores that differentially stain the viral membrane and its interior, we are able to monitor the kinetics of hemifusion and pore formation.
