Cell Culture Propagation

Cell culture propagation is the controlled expansion and maintenance of cells outside the body, enabling researchers to obtain sufficient, viable material for study and medical applications. Cells grow in a nutrient-containing culture medium under regulated conditions such as temperature, pH, gas exchange, and sterility; adherent cells are typically detached and reseeded during passaging, while suspension cells are diluted into fresh medium. This process supports disease modeling, drug screening, vaccine and biologic production, and studies of cell behavior. Careful monitoring of confluence, viability, contamination, and passage history helps produce reproducible cultures and reliable experimental results.

Propagation of Homalodisca coagulata virus-01 via Homalodisca vitripennis Cell Culture

Here we present a protocol to propagate Homalodisca vitripennis cells and HoCV-1 in vitro. Medium was removed from HoCV-1 positive cultures and RNA extracted every 24 hr for 168 hr. Cell survivability was quantified by trypan blue staining. Whole virus particles were extracted post-infection. Extracted RNA was quantified by qRT-PCR.

Limiting dilution cell transplantation assays are used to determine the frequency of tumor-propagating cells. This protocol describes a method for generating syngeneic zebrafish that develop fluorescently-labeled leukemia and details how to isolate and transplant these leukemia cells at limiting dilution into the peritoneal cavity of adult zebrafish.
