Cell Sorting Purity

Cell sorting purity is the proportion of collected cells that belong to the intended population after a separation procedure, making it a key measure of sample quality in medical research and diagnostics. In fluorescence-activated cell sorting, instruments identify cells by light-scattering properties and fluorescent markers, then use electronic gating and droplet deflection to separate target cells from unwanted populations. High purity reduces biological interference and improves the reliability of downstream analyses, including molecular profiling, functional assays, and cell-based therapies. Evaluating purity alongside recovery and viability helps researchers balance accurate population isolation with sufficient cell yield for clinical and laboratory applications.

The goal of this protocol is to isolate lymphatic endothelial cells lining human lymphatic malformation cyst-like vessels and foreskins using fluorescence-activated cell sorting (FACS). Subsequent cell culturing and expansion of these cells permits a new level of experimental sophistication for genetic, proteomic, functional and cell differentiation studies.

For many scientific studies requiring a biological and chemical analysis of cell populations the cells must be in a high state of purity. Fluorescence activated cell sorting (FACS) is a superior method in which to obtain pure cell populations.
