Cisplatin-induced Aki

Cisplatin-induced acute kidney injury (AKI) is a sudden decline in renal function caused by the anticancer drug cisplatin, and it is a major dose-limiting complication of chemotherapy. After filtration and uptake by proximal tubule cells, cisplatin forms DNA adducts and triggers oxidative stress, mitochondrial dysfunction, inflammation, and cell death, reducing tubular function. Understanding this mechanism supports serum creatinine monitoring, urine assessment, intravenous hydration, dose adjustment, and investigation of nephroprotective strategies and early biomarkers. Studying cisplatin-induced AKI also helps researchers evaluate renal toxicity and improve the safety of cancer treatment.

Acute Kidney Injury Model Induced by Cisplatin in Adult Zebrafish

This protocol describes the procedures to induce Acute Kidney Injury (AKI) in adult zebrafish using cisplatin as a nephrotoxic agent. We detailed the steps to evaluate the reproducibility of the technique and two techniques to analyze inflammation and cell death in the renal tissue, flow cytometry and TUNEL, respectively.

Early stage hemodynamic dysfunction is critical to the development of kidney disease. Yet, detection methodologies are limited. Recent advances in sonography provide a noninvasive, accurate option for early detection of kidney injury. This study outlines a step-by-step, sonographic methodology for detecting kidney dysfunction using a drug-induced nephrotoxicity rat model.

Here, we show the procedures for FAM83A knockdown; the assays to detect its effects on proliferation, migration, and invasion of cervical cancer cells; and the sensitization of these cells to cisplatin. This study provides a promising target gene for cervical cancer and a reference for further drug research.
