Fitc Inulin Clearance

FITC-inulin clearance is a fluorescence-based method for estimating glomerular filtration rate, a key measure of kidney function. After administration, fluorescein isothiocyanate (FITC)-labeled inulin circulates in the blood and is freely filtered by the glomeruli without significant tubular secretion, reabsorption, or metabolism; the rate of fluorescence disappearance from plasma or appearance in urine reflects clearance. This approach provides a sensitive, minimally invasive alternative to conventional inulin assays and can support assessment of renal function in experimental medicine. It is especially useful for detecting changes in filtration caused by kidney disease, nephrotoxic treatments, or physiological interventions.

This video demonstrates a method to study permeation through an in vitro blood-brain barrier (BBB) model using astrocytes and endothelial cells on a membrane insert, with free FITC and FITC-loaded ferritin. FITC-ferritin bound to receptors on endothelial cells and crossed the membrane barrier to the lower chamber while free FITC showed restricted permeation and remained in the upper chamber.
